Insight: New doubts about prostate-cancer vaccine Provenge

NEW YORK (Reuters) - Prostate cancer vaccine Provenge has long incited passions unlike any other cancer therapy.

Doctors who raised doubts about it received death threats. Health regulators and lawmakers faced loud protests at their offices. A physician at the American Cancer Society was so intimidated by Provenge partisans that he yanked a skeptical discussion of it from his blog.

The vitriol dissipated in April 2010, when the U.S. Food and Drug Administration approved Provenge for advanced prostate cancer, satisfying investors in manufacturer Dendreon and patients who for years had demanded it be put on the market.

But the bell on Round Two sounded when Marie Huber, a trained scientist and former hedge-fund analyst, made it her mission in the last year to analyze what she believes are deadly flaws in the studies that led to the approval of Provenge by the FDA.

She argues that the main reason Provenge seemed to extend survival - a crucial factor in the FDA's decision - was that older men in the study who did not receive Provenge died months sooner than similar patients in other studies.

She raises the possibility the "placebo" they received was actually harmful and made Provenge, known scientifically as sipuleucel-T, look better by comparison.

As Huber gains traction, most notably with a February paper in the prestigious Journal of the National Cancer Institute, she, too, is receiving threats. One post on an investors' message board last month suggested that "somebody smack her with a rubber hose." An email said "don't think you will be unscathed in this battle you waged on Provenge."

Provenge is Dendreon's only product and the company's stature with investors has waned with disappointing sales. In 2011, product revenues totaled $213.5 million, far from the $400 million Dendreon initially projected.

The company insists Huber's analysis is flawed and that Provenge has helped thousands of men with prostate cancer.

"I'm looking forward to getting this to patients around the world," said President and Chief Executive John Johnson.

FIRST CANCER VACCINE

Since it won FDA approval two years ago, Provenge has been Exhibit A for the idea that a patient's immune system can control or cure cancer. The first therapeutic cancer vaccine to reach the market, Provenge tries to engineer white blood cells, part of the immune system, to vanquish prostate cancer, which killed an estimated 33,720 men in the United States last year.

Its path to approval has all the features of a heavyweight healthcare fight - desperate patients demanding access to a promising therapy, a very expensive drug that extends life only a few months and efficacy data open to interpretation.

The FDA declined to approve the drug in 2007, when a clinical trial failed to show it slowed tumor growth. That incited protests, lawsuits and death threats against physicians on the FDA advisory panel who did not recommend approval, breaking with the 13-4 majority in favor.

"Provenge came along when we didn't have much to offer for prostate cancer," said Dr. Len Lichtenfeld of the ACS. "The advocacy community was bursting at the seams for something that worked. When you have that situation, it inflames passions and that can overtake the science."

In the pivotal trial called IMPACT, published in July 2010, but shared with the FDA months earlier, Provenge extended median survival by 4.1 months to 25.8 months from 21.7 months. That was sufficient for FDA approval. The vaccine costs $93,000 and patients also incur physician and other charges. Medicare agreed to cover Provenge last year, as have private insurers, but doctors initially balked at a long wait for reimbursement.

Huber had long been "utterly intrigued" by Provenge and its "huge promise of harnessing the immune system to battle cancer," she said in an interview.

In documents JNCI requires authors to sign, she declared no financial conflicts of interest. Neither she nor her former firm nor anyone else she is connected to stands to benefit financially from her analysis, she said.

Instead, she says she is motivated to help "vulnerable and desperate patients" - so much so that she gave up her job, salary and health insurance. Arguing that Provenge is harming these men, she called "the whole thing utterly horrific. The company got away with hiding data and doctors making $7,000 per prescription won't even engage in discussion" about whether it helps their patients.

After receiving degrees in biochemistry and bioscience enterprise from Cambridge University, Huber began working as an analyst for a hedge fund in 2007. A Thomson Reuters analysis of securities filings confirmed her former firm has not held any positions in Dendreon.

LACK OF EVIDENCE

Each dose of Provenge is custom-made. A nurse or technician withdraws white blood cells from a man's arm in a three-to-four hour procedure called leukapheresis.

The cells are shipped to a Dendreon manufacturing facility, where for two days they are incubated with a "fusion protein:" One protein that stimulates the cells' growth and maturation and another called PAP, or prostatic acid phosphatase. PAP is an antigen that studs prostate cancer cells like antennae, pieces of it sticking out of the cells' surfaces.

Dendreon says the patients' white blood cells take up the antigen and within hours their surfaces bristle with fragments of the telltale molecule. The cells are then shipped back to the physician and infused into the patient. A full treatment includes three such procedures, two weeks apart.

Back inside the body, Dendreon claims the modified cells trigger the immune system to produce T cells that kill any cell sporting the PAP antigen ? namely, prostate cancer cells.

In principle, that should eliminate the cancer, but Provenge does not shrink either the primary tumor or metastases.

Steven Rosenberg of the National Cancer Institute, a leading tumor immunologist, says that raises doubts over whether Provenge helps patients live longer, as the IMPACT trial reported.

"We have a lot of data that supports the idea that the product works the way it was designed to," said Dr. Mark Frohlich, Dendreon's chief medical officer. "We're seeing evidence of immune-system activation. The only question is whether the T cells are killing the tumor."

The FDA acknowledges that data supporting Provenge's approval did not show the drug shrank tumors, but says the overall survival benefit was enough to bring it to market. Spokeswoman Rita Chapelle, citing data submitted by Dendreon, said there is a "lack of evidence of anti-tumor activity," the reason for which "is unclear."

SUSPICIONS OVER SURVIVAL BENEFIT

Huber's analysis comes from data showing that men who received the placebo had very different survival times based on their age. Men older than 65 lived 17.3 months on placebo and 23 months with Provenge. Men younger than 65 lived 28 months after receiving placebo and 29 months after Provenge.

Other studies have shown that age generally does not affect how long a man survives with this form of prostate cancer, says Peter Iversen, a urologist and prostate-cancer surgeon at the University of Copenhagen and co-author of the paper with Huber.

Combining these findings led to the new paper's conclusion: The four-month edge in median survival from Provenge for all patients was due to longer survival among older men who got the vaccine.

"There is no efficacy in the younger patients, the primary group where you would expect it," said Huber.

Since the immune system weakens with age, an immune-based therapy should work better in younger men.

Some experts agree.

"If it was really a vaccine, you'd think younger men would show more response, since they are more immunocompetent," said NCI's Rosenberg.

On that basis, Huber and her co-authors, including two prostate-cancer specialists, argue the placebo used in the trial may have harmed the older men, cutting months off their lives and inadvertently making Provenge seem beneficial.

One way that could have occurred was through leukapheresis. That process removed about 90 percent of certain kinds of circulating white blood cells, according to calculations by immunologist Laura Haynes of the Trudeau Institute, a co-author of the JNCI paper.

The Provenge men got back about 32 percent of those cells, which had been stored at body temperature. The placebo men got back 12 percent, which had been incubated at near-freezing temperatures. Cold storage has been reported to kill "most, if not all, of those cells," notes the JNCI paper. Moreover, said Haynes, "if you return dead and dying cells to older men you are likely to cause inflammation," which can stoke the growth of cancerous cells.

Younger men were better able to replace the lost white blood cells, argued Iversen. Older men could not, resulting in early death.

"These cells are very specialized and there is research suggesting that removing them can harm older men," he said.

Earlier this month, DynaMed, an online database used by physicians, added to its Provenge entry a note on the JNCI paper, but calls the concern "not substantiated." ACS's Lichtenfeld says the analysis "might inhibit some patients and doctors from going ahead with a very expensive drug."

Huber says she plans to approach European regulators as they consider Dendreon's application to approve Provenge.

Investor message boards have lit up in response to the new paper. In February, an anonymous commentator on InvestorVillage.com warned that Huber's work was about to be published "a few days before our earnings. Her agenda is obvious."

IFFY STATISTICS

Critics of the new analysis argue the number of cells removed is too small to suppress the immune system. Charles Drake, an oncologist and immunologist at Johns Hopkins Sidney Kimmel Comprehensive Cancer Center, said there is no evidence the placebo men in IMPACT suffered more infections or other effects of a depleted immune system than the Provenge men.

The scientist who led IMPACT, oncologist Philip Kantoff of Dana Farber Cancer Center, said colleagues in immunology "dismissed as nonsense the idea that leukapheresis could hurt individuals."

He takes issue, too, with the statistics. Dividing the men by whether they are older or younger than 65, he said, is "arbitrary" and to pick apart data retrospectively is a statistical no-no.

Dendreon's Frohlich also criticizes the statistics: "If you do enough of these (post-hoc analyses) then by chance alone you'd expect to get one positive finding."

In other words, it is almost always possible to find a subset of patients who do better than others.

When Dendreon divided the men by whether they were older or younger than about 71, he added, they found no red flags.

The FDA agrees that such post-hoc statistical analyses "are exploratory" and their results "must be interpreted with caution, as acknowledged by the authors." Yet a paid consultant to Dendreon before the IMPACT trial agreed with many of Huber's concerns.

"The control vaccine used in IMPACT and in the predecessor trial had never been used anywhere for anything and may well have been detrimental to patients," said Donald Berry of MD Anderson Cancer Center, a leading biostatistician. "Here's a great way to get your drug approved: Kill the control patients."

Despite the heated rhetoric, Provenge may go out with a whimper more than a bang. Promising new agents for advanced prostate cancer include an oral drug from Medivation Inc called enzalutamide, in the final phase of clinical trials, and Zytiga from Johnson & Johnson, which won FDA approval in 2011. A vaccine that targets PSA, from Bavarian Nordic Immunotherapy, is in late-stage trials.

Dendreon does not disclose how many patients have been prescribed Provenge. CEO Johnson said that about 70 percent of its Provenge revenue comes from sales to community hospitals and doctors and 30 percent from academic medical centers. Some of the latter decline to use Provenge, deterred by lingering concerns over whether it provides a meaningful benefit.

Three such facilities in the Midwest, contacted at random by Reuters, confirmed they do not recommend Provenge. All asked not to be named for fear of receiving threats.

"It is my policy not to make public comments about this drug," said one oncologist. Patients who ask for it "are referred to another facility."

Spike Lee, latest celeb to practice reckless tweeting

Spike Lee wasn't the first to retweet the erroneous address on Twitter blindly accepted by many as that of George Zimmerman, the man who fatally shot 17-year-old Trayvon Martin last month. But the filmmaker's message -- sent last week to his 240,000 Twitter followers -- was the most viral.

Elaine McClain, 70, the owner of the misidentified house in Sanford, Florida, and her husband David McClain, 72, tell news outlets they're now "living in fear." The address spread on Twitter, accompanied by threats of violence, and the couple are now staying at a hotel "to avoid the spotlight and possible danger," reports the Orlando Sentinel.

Oh, and Mrs. McClain? She reportedly has a heart condition. Awesome.

So how did this happen? As The Smoking Gun breaks it down, Marcus Davonne Higgins, the 33-year-old Los Angeles man behind the bad address, confused McClain's son, George W. Zimmerman, who once lived at the address, with George Michael Zimmerman, the man who killed Martin. Higgins then blasted the address to the Twitter accounts of Will Smith, 50 Cent, LeBron James, as well as to Lee. But Lee is the only one who retweeted it.

The great big "WRONG!" here isn't the bad address, or whether Lee should research the info before he retweets. And as regrettable as it is, Lee can't be held accountable for giving conservative pundit Michelle Malkin more fuel for Trayvon Martin race baiting on her Twitter "news" site Twitchy. It's this: Celebrities have no business using their position of power and popularity on Twitter to incite a mob.

We decry it when members of 4chan's /b/board broadcast the phone number, address and other identifying info about young girls suddenly popular on the Internet. Here in the West, we're horrified by stories of China's Internet mob, "mobilized to root out, expose and shame people they perceive to be exhibiting corrupt or immoral behavior," sometimes resulting in real-world violence. It's equally not OK for U.S. celebrities to do the same.?

We saw this to a lesser degree last week when Alec Baldwin mustered a Twitter army against someone he claimed was harassing his girlfriend on the Internet. When a celebrity directs his fanatic legion against a person outside a position of power, that's straight-up bullying.

Posting on Twitter, Baldwin gave out the handle of the alleged?harasser, @scarfacemadam, whom he said "has entered the twitter-verse ONLY to harass my girlfriend" in a way "that is borderline threatening." He asked users to "go to her address and read just a few of her posts, and give her your notice of her behavior."

While many of the Twitter attacks against?@scarfacemadam didn't?compare to the jillions of ugly death threats tweeted by Justin Bieber fans on any given day, they weren't pretty. As we noted then,?if the troll harassing his GF really is as bad as he claimed, then it's way to fuel a psycho stalker, Alec Baldwin! Conversely, if the troll was just your run-of-the-mill Internet jerk (with or without diagnosable mental issues), then Baldwin just sent thousands of people in that troll's direction.?

As it turned out, the kerfuffle faded fast soon after the @scarfacemadam account went dormant. It could've been worse. It wasn't.

In Lee's case however, helping chum Twitter could've resulted in violence -- against an innocent elderly couple or a man yet to face the justice system -? as well as a jail sentence for any Lee fans who took his retweet to heart. ?

Remember the so-called Mozart Effect on babies? intelligence? Well, that pseudo-science or pop psychology seems recently to have been pretty well debunked and discredited.

But what about animals and music?

For many years, I have sworn that my cat Rosie (below) loves music, just as I do, especially piano music.

Rosie is a sweet and pretty tabby cat, and she seems to come over by the piano and sit down or lie down and roll over, or even jump onto my lap hile I am playing whenever I start practicing.

It seems to happen especially whenever I am playing Bach, Schubert or Chopin.

So I wondered: Is it me and the fact she identifies the piano sound with my presence, the same way Pavlov?s dogs responded to bells? Or is it the music?

Well, it is probably some of each, says Charles T. Snowden, a University of Wisconsin-Madison researcher and animal psychologist.

Among his findings are that animals show some breed specificity in the music they prefer. That is, they follow their own species? taste or preference rather than their owner?s taste or preference. More than a decade before former Penn State coach Jerry Sandusky was charged with more than 50 counts of child sex abuse, a psychologist warned university police that his actions fit that of a ?likely pedophile?s pattern.?

The finding by State College, Pa., psychologist Dr. Alycia A. Chambers, the therapist for one of Sandusky?s alleged victims, was contained in the internal Penn State files of a 1998 police investigation of the former coach for showering and bear hugging her client and another young boy in the school?s athletic locker room.

The Sandusky Files: Read the 1998 police report

NBC News has obtained the complete file on the investigation ? the police report and assessments by two psychologists who interviewed the boys -- which provides new details about Sandusky?s behavior. It also could raise fresh questions about how school and local authorities handled his case.

?There was very little doubt in my mind (Sandusky) ? was a male predator, someone that was in the process of grooming a young man for abuse ,? said Chambers, speaking publicly for the first time, with the permission of her client?s family, in an interview with NBC News. ?I thought?my report was strong enough to suggest that this was somebody who should be watched.?

The Sandusky Files: Child psychologist's report

Chambers? detailed report is potentially significant because it was the first clear warning about the former Penn State coach?s actions ? nearly four years before a then-graduate assistant, Mike McQueary, reported to the late Coach Joe Paterno and other top school officials that he had found Sandusky in the Penn State showers one evening with another young boy, engaged in what he viewed as sexual contact. (Paterno testified last year he was unaware of the 1998 investigation and Gary Schultz, the former Penn State vice president who oversaw the school police, testified that he never reviewed the details of the case. A Penn State spokesman declined comment, citing pending investigations.)

In her interview with NBC News, Chambers described her anguish when she was contacted by police last year and learned that authorities were again investigating Sandusky for allegedly molesting multiple other boys, 13 years after she first raised her concerns.

?I was horrified to know that there were so many other innocent boys who had their hearts and minds confused, their bodies violated,? said Chambers. ?It?s unspeakable.?

Statement from Joe Amendola, Sandusky's lawyer (.PDF)

Joe Amendola, Sandusky?s lawyer, said he hasn?t seen Chambers? report, but that her conclusions will be disputed by other psychologists who will be called by the defense. ?I understand that there are some people who could look at this behavior and say it?s a pedophile problem. But there are others who will say, ?This is somebody who loves kids and loves to be around them? ? It?s the old story, you get your expert and I?ll get my expert.? Sandusky has pleaded not guilty to all charges.

But one of the investigators on the 1998 case, Jerry Lauro, then with the state Department of Public Welfare and now retired, told NBC News he was never shown a copy of Chambers? report and was stunned to learn of its conclusions.

?Wow!? he said when he was read Chambers? conclusions by a NBC News correspondent. ?This is the first I?ve heard of this. I had no idea . If I would have seen the report, I would certainly have done some things differently. Boy, this is a shock. ?

Story: Lawyer seeks dismissal of charges against Sandusky

Chambers was the psychologist for a then-11- year-old boy, who had met Sandusky through his Second Mile charity for troubled children and was later invited by the defensive coordinator to several Penn State football games. The boy is now known as Victim 6?one of 10 allegedly molested by Sandusky and one of the key figures in the criminal case against him slated to go to trial this spring. Amendola this week asked a judge to drop Victim 6 from the case on the grounds that his anticipated testimony ?will not establish that any sexual contact took place.?

The police file provides a fuller picture than has been previously made public about Sandusky?s interactions with the boy as well as the transcripts of two tense confrontations between Sandusky and the boy?s mother.
Chambers was called by the boy?s mother at 7:43 a.m. on May 4, 1998. The boy had returned home the night before, his hair wet, after spending two hours with Sandusky at the Penn State athletic room.
After meeting with her client in her office and talking to his mother, Chambers described in her written report how Sandusky had coaxed the boy into the shower after a workout, telling him, ?All the guys do.? He then moved closer to him, squeezing him tightly from behind while they were both naked. The boy also told Chambers how, during their workout, Sandusky had kissed him on the forehead and told him, ?I love you.? He also invited the boy back to his house to ?sit on his lap? and go ?online? on his ?cool computer,? the boy?s mother told Chambers, recounting her conversation with her son the previous night.

Story: Penn State offers therapy to alleged abuse victims

?My consultants agree that the incidents meet all of our definitions, based on experience and education, of a likely pedophile?s pattern of building trust and gradual introduction of physical touch, within a context of a ?loving,? ?special? relationship,? Chambers wrote in her report.

?One colleague, who has contact with the Second Mile, confirms that Mr. Sandusky is reasonably intelligent and thus, could hardly have failed to understand the way his behavior would be interpreted, if known,? Chambers continued. ?His position at the Second Mile and his interest in abused boys would suggest that he was likely to have had knowledge with regard to child abuse and might even recognize this behavior as a typical pedophile ?overture.??

Chambers gave her report to Penn State police Officer Ronald Schreffler on May 7, 1998, along with a cover letter that highlighted ?the gravity of the incidents.? Chambers had also reported the incident to the Pennsylvania ?suspected child abuse? hotline, where officials wrote up their own report identifying Sandusky as the ?AP? or ?Alleged Perpetrator.? Lauro, an investigator who specialized in abuse cases, was assigned to work the case with Schreffler.

Investigators brought in a second psychologist, John Seasock, who had worked with the local Centre County Child and Youth Services, a local agency that had licensed Sandusky as a foster parent. After meeting with the boy for an hour, Seasock concluded that no sexual offense had taken place nor was there ?grooming? or ?inappropriate sexual behavior? by Sandusky.

Report of expert who concluded no abuse occurred

?All the interactions reported by (the boy) can be typically defined as normal between a healthy adult and a young adolescent male,? Seasock wrote in his report.

Lawyer: Report helps defense
Amendola, Sandusky?s lawyer, has said that he hopes to use Seasock?s report in his client?s defense. But the police files show that Seasock conducted his interview of the boy ?cold,? without reviewing Chambers? report or prior transcripts of interviews with the boy and that he failed to elicit some key details, such as Sandusky kissing the boy and telling him, ?I love you.?

Seasock acknowledged to police there were some ?gray areas? and that investigators ?can?t walk away from the investigation.? But he also appeared to dismiss most of the concerns that had been raised about Sandusky in an interview with investigators.

?SEASOCK said that he hasn?t heard of a 52-year-old becoming a pedophile,? the police report states. ?SEASOCK said that SANDUSKY didn?t fit the profile of a pedophile.? (Asked by police if it was "possible" that a man that age could become a pedophile, he replied it was "possible.?)

Seasock did not return phone calls from NBC News seeking comment. Schreffler also declined comment, saying only, ?The report speaks for itself.?

The Penn State police file shows that investigators continued to pursue the case under the supervision of the late Ray Gricar, then the local district attorney. According to the police report, they interviewed a second boy ? a friend of Victim 6 ? who also described being ?uncomfortable? when Sandusky hugged him in the Penn State showers.

In the days following Chambers? interview with the boy, the mother became concerned because Sandusky was continuing to call her son at his apartment, coming by the back of the family?s apartment with rear sliding doors and even showing up at a ball field where her son had Little League practice.
As Sandusky continued to try to contact the boy, the police hid in the bedroom of the family?s apartment and monitored two confrontations between the mother and Sandusky.

On the afternoon of May 13, 1998, Sandusky knocked on the door of the family home, looking for the boy. The mother greeted him instead, telling him ever since her son had seen him the previous week, ?he?s been acting different. He won?t talk and he?s been having nightmares.?

When Sandusky asked to talk to her son, the mother replied: ?I don?t want you calling here for (her son.) I need to think about this. Let me call you if it?s OK to pick (her son) up.

?Maybe I worked him too hard,? Sandusky replied.

?Did something happen?? the boy?s mother asked.

?I don?t think so,? replied Sandusky.

On May 19, police arranged to monitor a second meeting that turned even more tense and resulted in Sandusky having ?tears in his eyes,? according to the mother?s account to investigators.

The mother: ?Did your private parts touch (the boy) when you bear hugged him?
Sandusky: ?I don?t think so. ? Maybe.?


The mother: ?Have you ever done this with other boys??


Sandusky: ?Yes.?
The mother recounted how ?you told him you loved him and kissed him on the head.? Sandusky replied that the boy ?told me he loved me, so I told him the same thing.?


The mother said her son had looked at Sandusky ?as a hero, a father figure, he is really confused.?
Sandusky then pressed to talk to the boy again, but the mother told him that would not be a ?good idea.?
?I don?t want you going to (his) ball games.?


?I understand,? Sandusky replied. ?I was wrong. I wish I could get forgiveness. I know I won?t get it from you. I wish I were dead.?


Just a week and a half later, the two lead investigators ?Schreffler and Lauro?interviewed Sandusky at his office at the Penn State athletic building. He acknowledged that he hugged the boy in the shower ?but that there wasn?t anything sexual about it.? He also said that he had showered with other boys and ?realized he used poor judgment in what he did.?

?As a result of the investigation, it could not be determined that a sexual assault occurred and Sandusky was advised of such, ?the police report reads. One of the investigators then ?advised SANDUSKY not to shower with any child. SANDUSKY stated that he wouldn?t.?

?CASE CLOSED,? the file reads.

There are no reasons given in the report for why Gricar ultimately chose not to prosecute Sandusky based on the 1998 investigation. (In a case that remains open, Gricar disappeared in 2005 and is now presumed dead. Investigators say there is no indication his disappearance relates to Sandusky.)

In an in an interview with NBC News, the boy?s mother described telling her son that night that no charges would be brought against Sandusky.

Her son, she said, was ?confused.?

?I don?t understand, Mommy,? she said he replied. ?I?m just a little kid. I knew what he did was wrong. Why didn?t he (know it was wrong)??

Sandusky retired from Penn State the following year ? to spend more time working with Second Mile, he said. But he was made a ?professor emeritus? and maintained keys to the Penn State locker room, where he continued to take young boys to the showers. In a telephone interview Friday, Amendola was asked why Sandusky still showered with young boys after 1998 when police had told him not to ?and he told them he wouldn?t.

?He viewed that more as an admonition, not an order,? Amendola said. ?He didn?t think there was anything wrong with it.? Amendola added that Sandusky ?never thought that the situation in ?98 was a big deal. He never looked at as that serious.?

?It?s very disturbing,? said Walter Cohen, a former Pennsylvania attorney general in the mid-1990?s, who reviewed parts of the Penn State police file, including Chambers? report, at NBC News request. ?It never should have been ignored. Never.?

As a former secretary of public welfare in the 1980s, Cohen had set up a statewide ?child-line? registry of suspected child abusers that could serve as a central data base for school officials and others working with children to conduct background checks on prospective employees. Even if there were no criminal charges brought, Cohen said, there was more than enough evidence to have placed Sandusky on the statewide registry.

?Jerry Sandusky should have been on the watch list,? he said. ?But instead, the case was closed.?

BEIRUT (AP) ? Tens of thousands of Syrians braved tear gas and gunfire to protest across the country Friday, vowing to storm the capital Damascus to oust President Bashar Assad as the European Union ramped up pressure on the regime by imposing sanctions on his wife and other close relatives.

Security forces deployed in many cities to disperse protests, but opposition groups reported fewer protester deaths than in past weeks. Activists said more than 20 people were killed nationwide in army attacks on opposition areas or clashes with armed rebels.

International condemnation and high-level diplomacy have failed to stop the year-old Syria crisis, which the U.N. says has killed more than 8,000 people, many of them civilian protesters.

Friday's sanctions bring to 13 the sets imposed by the EU to try to compel the regime to halt its violent crackdown on dissent. The U.S. and others have also imposed sanctions. Previous measures were aimed at Syrian companies and Assad himself.

Those imposed Friday targeted Asma Assad, Syria's British-born first lady, banning her from traveling to EU countries and freezing any assets she may have there. They also included the president's mother, sister, sister-in-law and eight government ministers.

French Foreign Minister Alain Juppe said sanctions were weakening the regime.

"Their economic situation becomes ever more difficult. Syria has few reserves," he said. "We think its economic situation will become untenable."

While the measures have hurt Syria's economy, they appear to have had little effect on the regime's actions. It has regularly deployed troops, pro-government thugs and snipers to attack anti-regime protests. Human rights groups accuse the regime of shelling civilian areas and torturing and killing detainees in its push to stop the uprising, which it blames on terrorists carrying out a foreign conspiracy.

In Geneva, the U.N. Human Rights Council blasted Syria's crackdown and extended the mandate of a U.N. expert panel tasked with reporting on alleged abuses in the country.

A resolution passed by the 47-member body condemned "widespread, systematic and gross violations of human rights and fundamental freedoms perpetrated" by Syrian authorities, including summary executions, torture and sexual abuse of detainees and children.

Also Friday, UNICEF said at least 500 children have been killed in the conflict, while hundreds more have been injured, detained or abused. The U.N. children's agency said schools have closed and health centers have shut down or become too dangerous for many families to reach.

Throughout the conflict, China and Russia have protected Syria from censure by the U.N. Security Council, fearing a strongly worded resolution condemning Assad could pave the way for military intervention, as happened in Libya last year.

Russia, however, softened its stance Thursday by calling for Assad to pull his troops out of Syrian cities. The U.N. has been trying to secure a cease-fire so all parties could hold a dialogue on a political solution to end the conflict. So far, both sides have refused talks.

Regime forces continued to pound oppositions areas Friday, and activists reported major shelling and fire with heavy machine-guns in the provinces of Homs in central Syria, Idlib in the north and Daraa.

The London-based Syrian Observatory for Human Rights said at least 23 civilians were killed in government attacks Friday. Government troops and armed rebels clashed in a number of places, with at 13 soldiers and three rebel fighter killed, the group said.

Another group, the Local Coordination Committees, said government troops killed 36 civilians on Friday. It did not provide details on each civilian killed.

Activists reported dozens of anti-regime protests in towns and cities across Syria under the banner "Damascus, we are coming." Security forces broke up many of them with gunfire and tear gas, and there were reports of wounded.

Activists reported fewer protester deaths and Rami Abdul-Rahman, head of the Observatory, said he had yet to confirm a single protester death on Friday, remarking that this was unusual.

"We hope it happens like this every time because we don't want anyone to die," he said.

The Syrian government has barred most media from working in the country, and activist accounts could not be independently verified.

Syria's state news agency said hundreds marched in a pro-Assad demonstration in the capital Damascus and published photos of them carrying Syrian flags and Assad photos.

In Jordan's capital Amman, blind Syrian cleric Ahmad al-Sayasneh called on a congregation of 1,000 Syrians to "remain steadfast until our tyrant leadership is ousted."

It was the cleric's first public appearance since fleeing Syria two months ago. Al-Sayasneh rose to prominence though his fiery sermons calling for civil disobedience at a mosque in the southern Syrian town of Daraa, considered the uprising's birthplace.

Instagram opens signup page for Android port, release date still unknown

It's no secret that one of the most popular apps to ever hit the App Store will soon be coming to Android, and if you'd prefer to be one of the very first on your block to be notified... well, there's a website for that. Instagram has just opened up a signup page for Android loyalists, enabling folks to input their email address and await word on the download going live. Sadly, there's no hint on the aforesaid page that gets us any closer to a specific release date, but hey -- it's one less unspecified thing you have to remember, right?
